Проблема выявления бактериальных липополисахаридов (ЛПС) в составе фармацевтических субстанций и реагентов сохраняет свою актуальность. Относительно недавно было показано, что TLR4-PTEN/PI3K/AKT/NF-κB-сигнальный путь принимает участие в нейровоспалении в нейронах гиппокампа крысы после воздействия ЛПС E. coli. Можно сделать вывод, что существует связь между количеством ЛПС и его воздействием на синтез провоспалительных цитокинов / хемокинов и факторов роста в нейронах человека. Для подтверждения теории и поиска изменения уровня цитокинов / хемокинов на уровне белков в нашей работе была выбрана клеточная культура нейробластомы человека линии SH-SY5Y как одна из наиболее изученных нейрональных моделей.</p><p>Цель исследования – анализ секретома (цитокинов / хемокинов и факторов роста), участвующего в запуске синтеза цитокинов в нейронах из культуры нейробластомы SH-SY5Y после внесения ЛПС Salmonella typhosa.</p></sec><sec><title>Материал и методы</title><p>Материал и методы. Для определения содержания цитокинов / хемокинов и факторов роста в клетках культуры нейробластомы SH-SY5Y после воздействия ЛПС использовали многопараметрический иммунофлуоресцентный метод. Потенциальные маркеры определяли методами статистического анализа.</p></sec><sec><title>Результаты</title><p>Результаты. Воздействие ЛПС из Salmonella typhosa на культуру клеток нейробластомы SH-SY5Y привело к изменениям в секреции цитокинов / хемокинов на уровне белков в период с первого по четвертый день после введения ЛПС.</p></sec><sec><title>Ограничения исследования</title><p>Ограничения исследования. Исследование in vitro на культуре клеток показало зависимость цитокинового / хемокинового секретома от исследуемых соединений, но результаты нельзя экстраполировать на живой организм.</p></sec><sec><title>Заключение</title><p>Заключение. Наличие дозозависимых эффектов воздействия бактериальных эндотоксинов на клеточные культуры позволяет рассматривать «определение уровней маркерных цитокинов» в качестве возможного метода для количественного анализа уровня ЛПС.</p><p>Соблюдение этических стандартов. The problem of identifying bacterial lipopolysaccharides (LPS) in pharmaceutical substances and reagents remains relevant. recently, it was shown that the TLR4-PTEN/PI3K/AKT/NF-kB signaling pathway participates in neuroinflammation in rat hippocampal neurons after exposure to i LPS. It can be concluded that there is a relationship between the amount of LPS and its effect on the synthesis of pro-inflammatory cytokines, chemokines, and growth factors in human neurons. To confirm the theory and to search for changes in cytokine/chemokines levels at the protein level, we selected the SH-SY5Y human neuroblastoma cell culture as one of the most studied neural models.</p><p>The aim of the study was to analyze the secretome (cytokines / chemokines, and growth factors) involved in triggering cytokine synthesis in neurons from the culture of neuroblastoma SH-SY5Y after the action of lipopolysaccharides Salmonella typhosa.</p></sec><sec><title>Material and methods</title><p>Material and methods. To evaluate the concentrations of cytokines / chemokines, and growth factors in SH-SY5Y neuroblastoma culture cells after exposure to LPS, we utilized a multi-parameter immunofluorescence method. Potential biomarkers were identified through statistical data analysis methods.</p></sec><sec><title>Results</title><p>Results. The effect of LPS from Salmonella typhosa on neuroblastoma SH-SY5Y cell culture led to changes in cytokine / chemokine secretion at the protein level from the first to the fourth day after LPS administration.</p></sec><sec><title>Limitations</title><p>Limitations. An in vitro study on cell culture has shown the dependence of the cytokine/chemokine secretome on the studied compounds, but the results cannot be extrapolated to a living organism.</p></sec><sec><title>Conclusion</title><p>Conclusion.
