Уточнение токсичности мефлохина и оценка эффективности его применения при заражении сирийских золотистых хомяков SARS-CoV-2.</p></sec><sec><title>Материал и методы</title><p>Материал и методы. Эксперимент проводили на 96 сирийских золотистых хомяках. Определяли токсичность при однократном введении, а также при курсовом введении в дозах, сопоставимых с терапевтическими для человека. Для изучения эффективности при заражении SARS-CoV-2 использовали комплексный показатель состояния лёгочной ткани и сравнение динамики вирусной нагрузки.</p></sec><sec><title>Результаты</title><p>Результаты. Среднелетальная доза мефлохина при однократном пероральном введении составляет 817 мг/кг, максимально переносимая доза — 600 мг/кг. При введении курсом 7 дней в кумулятивной дозе 900 мг/кг гибели животных не отмечалось. Введение мефлохина животным на фоне заражения SARS-CoV-2 сопровождалось уменьшением тяжести поражения лёгких и более быстрым снижением титра вируса в лёгочной ткани.</p></sec></abstract><trans-abstract xml:lang="en"><p>The aim of the study was to clarify the toxicity of mefloquine and to evaluate the effectiveness of its use for Syrian hamsters infected with sARS-Cov-2.</p><sec><title>Material and methods</title><p>Material and methods. The experiments were performed on 96 Syrian hamsters. The toxicity of mefloquine was determined with a single administration, as well as with a course of administration at doses comparable to therapeutic for humans. To study the effectiveness of the drug against SARS-CoV-2 infection, a comprehensive indicator of the state of lung tissue and a comparison of the dynamics of viral load in the lungs were used.</p></sec><sec><title>Results</title><p>Results. The LD50 of mefloquine with a single oral administration is 817 mg/kg, the maximum tolerated dose is 600 mg/kg. When administered for 7 days at a cumulative dose of 900 mg/kg, no death was observed.
